Immunomodulatory Activity of Platycodon grandiflorum, Codonopsis lanceolata, and Adenophora triphylla Extracts in Macrophage Cells

|
This study investigated the immunomodulatory and anti-inflammatory activities of water extract of Platycodon grandiflorum, Codonopsis lanceolata, and Adenophora triphylla (PG, CL, AT) in a murine RAW264.7 macrophage model. As immuno response biomarkers, the cell proliferation, nitric oxide (NO) production, tumor necrosis factor (TNF)-¥á production and degree of mitogen-activated protein kinase (MAPKs) phosphorylation in RAW264.7 macrophage cells were evaluated. PG and CL increased macrophage proliferation and production of NO and TNF-¥á, with greater immunostimulatory activities being observed in response to PG than CL in RAW264.7 cells. The increase of NO and TNF-¥á in PG or CL-treated cells was associated with phosphorylation of MAPKs, including extracellular signal-regulated kinase (ERK), p38 and c-Jun N-terminal kinase (JNK) in RAW264.7 cells. The phosphorylation of ERK, p38 and JNK (p-ERK, p-p38, p-JNK) of RAW264.7 cells treated with PG and CL, respectively, also increased significantly. The NO and TNF-¥á production and phosphorylation of ERK and p38 were unchanged in AT-treated RAW264.7 macrophages. Furthermore, we examined the effects of PG, CL and AT on NO and TNF-¥á production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. PG, CL and AT inhibited LPS-induced NO and TNF-¥á production. These findings suggest that PG and CL can induce macrophage activation through MAPK signaling as immunomodulators and that they exert anti-inflammatory effects through inhibition of NO and TNF-¥á production.
